GIST patients show good response rates to new drug

Those with gastrointestinal stromal tumors (GIST) resistant to imatinib (Gleevec) may benefit from a new drug according to the results from a phase II study.

More than half of patients develop resistance to Imatinab, which is currently the first-line therapy.

The experimental drug, code named SU11248, showed response rates in 60 of the 92 patients treated who were resistant to imatinab. SU11248 blocks multiple pathways involving tyrosine kinase, VEGF, PDGF, and the KIT receptor. Patients with mutations in KIT exon 9 showed the strongest response. SU11248 had a safety profile similar to that of imatinib, such as fatigue, nausea, diarrhea, gastrointestinal bleeding, and an increase in blood pressure in some patients, probably due to the drug's angiogenic effects.
